The Ferry left Nusa Penida at 0830 and arrived at Padangbai at ~ 10am then "turned round" and departed Padangbai at 11am and arrived Penida at 1230 - only one Service. There is a single loket which sells tickets. It leaves about am. If you miss this this boat, the RoRo ferry from the main wharf takes foot passengers and leaves between am to am and between pm to pm.
